I know sedistain is the recommended stain to use but my question is does it matter and why? I saw a vet using diff-quick

RE: Staining

Thank you to you all this is really helpful!!:-)

RE: Staining

Kova is a lot cheaper than sedistain - does the same job, yes I used to split larger bottles of stain into smaller quantities - especially if I had a bunch of students, it helps a lot to cut down on contamination - the small centrifuge tubes are ideal.

methylene blue (which is as near as dammit your solution C) is actually pretty good for staining urine preps

RE: Staining

Hi - I wouldn't say that diff-quick is 'better' - it's just an alternative method. You need to make a dry prep of the urine (smear the sediment and wait for it to dry) but I guess you saw the vet do that. I'm not a great fan of sedi-stain myself, I find it tends to stain EVERYTHING and makes it harder to pick out what you might be trying to find. In addition, the sedi stain pot has the potential to be a breeding ground for bugs, which could then be wrongly associated with the urine (I think it was Sal's suggestion to decant the bottle of sedi-stain into smaller containers to avoid this issue)

I like to make an unstained prep and a stained prep on the same slide, then you can look at both. Try making some diff-quick preps too and see which you prefer.
